This talk addresses key preclinical and early translational challenges in oncology model-informed drug development. It explores emerging immunomodulatory therapies,alongside the rational design of combination strategies to enhance efficacy. The optimization of treatment sequencing and its impact on biological response will also be discussed. Emphasis will be placed on the role of biomarkers for stratification and response assessment across model systems. Finally, challenges related to drug distribution and tumor penetration will be examined, highlighting their importance in achieving adequate target engagement and supporting early decision-making in oncology drug development.
